Description: This R2.477 monoclonal antibody reacts with human CD112 (also known as Nectin-2 or Poliovirus Receptor Related 2), a 60-65 kDa immunoglobulin-like transmembrane cell adhesion molecule. This cell surface molecule is expressed on hematopoietic cells, endothelial, epithelial, as well as tumor cell lines such as Jurkat, U937, and MCF7. CD112 is a ligand for DNAM-1 (CD226), a leukocyte adhesion molecule expressed on natural killer (NK) cells. Interaction between CD112 and DNAM-1 promotes NK cytotoxic activity, leading to tumor cell lysis. In addition to its role in tumor suppression, CD112 plays a cooperative role in cell-cell adhesion, particularly in the formation of adherens junctions between neighboring epithelial cells and fibroblasts. CD112 can form homophilic and heterophilic interactions to regulate contact inhibition of cell movement and proliferation. Finally, CD112 has been reported to be a receptor for herpesvirus entry into cells.
This monoclonal antibody has been reported to block cell adhesion and herpesvirus entry.
Applications Reported: This R2.477 antibody has been reported for use in flow cytometric analysis.
Applications Tested: This R2.477 antibody has been pre-diluted and tested by flow cytometric analysis of normal human peripheral blood cells. This may be used at 5 µL (0.25 µg) per test. A test is defined as the amount (µg) of antibody that will stain a cell sample in a final volume of 100 µL. Cell number should be determined empirically but can range from 10^5 to 10^8 cells/test.
